Recently, there have been continuous updates on the progress of domestic pharmaceutical companies in the field of antibody-drug conjugates (ADCs). On December 5th, Hengrui Medicine announced that the indication for its antibody-drug conjugate SHR-A2102 for urothelial cancer has been included in the list of proposed breakthrough therapy products. Not long ago, KaryoBiotec and Huadong Medicine have successively announced that their ADC drugs have been approved for marketing in China. What is the overall target landscape of domestic pharmaceutical companies in the ADC drug field? Which targets are more promising? How do you view the future development trend of the domestic ADC drug market? Looking globally, what is the growth potential of the ADC drug market?

 

Wang Peng, project manager of Frost & Sullivan's Life Sciences division in Greater China, was interviewed by China Securities Journal to discuss the current development status, market prospects, and future competitive landscape of the ADC field both domestically and internationally.

 

Q:On November 27th, KaryoBiotec announced that its rucaparib monotherapy has been approved for marketing, becoming China's first domestically developed innovative TROP2 ADC. Could you introduce the overall target landscape of domestic pharmaceutical companies in the ADC drug field and which targets are more promising?

Compared to the global ADC clinical research targets, domestic pharmaceutical companies' target landscape in the ADC drug field is relatively concentrated. Popular research targets include well-established targets such as HER2, TROP2, EGFR, and CLDN-18.2. Domestic pharmaceutical companies usually adopt methods such as optimizing drug design and differentiated indications to find breakthroughs in the fierce competition. At the same time, since targets are an important innovation area of ADC drugs, with the development and progress of multi-omics technology in recent years, domestic enterprises have continuously made breakthroughs and innovations in the ADC drug research and development field through the layout of new targets. Currently, the target selection of ADC drugs has expanded from traditional antigens expressed on the surface of tumor cells to the tumor microenvironment, showing potential for developing new ADC drugs.

 

 

Q:In the recently released 2024 National Medical Insurance Catalogue, two ADC drugs, Roche's vedotuzumab for injection and trastuzumab for injection jointly developed by First Tropix and AstraZeneca, have entered the medical insurance catalog for the first time. How do you view the future development trend of the domestic ADC drug market? Looking globally, what is the growth potential of the ADC drug market?

The inclusion of vedotuzumab and trastuzumab in medical insurance not only reduces the economic burden on patients and benefits a large number of domestic patients but also marks an important progress in the market access of ADC drugs in China, which will further drive the development of the domestic ADC drug market. According to Frost & Sullivan's data analysis, the global ADC drug market size has always maintained high-speed growth, increasing from $2 billion in 2018 to $10.4 billion in 2023, with a compound annual growth rate of 39.1%; it will continue to grow at a compound annual growth rate of 30.3% from 2023 to 2030, and it is expected that the global ADC drug market size will grow to $662 billion by 2030.

 

 

Q:As more and more domestic pharmaceutical companies enter the ADC field, market competition will become more intense, putting forward higher requirements for product differentiation and market positioning. Which enterprises with future advantages may emerge victorious in the competition?

ADC drugs have revolutionized the treatment paradigm of cancer, bringing huge market opportunities. While attracting a large number of innovative pharmaceutical companies and capital, they also test the R&D innovation, clinical design, strategic execution, and commercialization capabilities of major pharmaceutical companies. Innovation is the driving force for breakthroughs in ADC drugs. Although the design concept of ADC drugs seems simple, due to its three-part combination, the complex and variable components make the R&D innovation of ADCs more challenging for pharmaceutical companies' combined innovation capabilities. At the same time, the ultimate manifestation of the innovative value of ADC drugs is in clinical results. One of the important factors for the successful development of ADC drugs lies in establishing effective clinical trials and treatment plans. In addition, commercial production is the key point for the transformation of ADC drug innovation achievements. From drug discovery to the commercial development of a single ADC product, it usually requires a large amount of continuous capital investment, ranging from hundreds of millions to over a billion dollars. Therefore, enterprises with comprehensive competitive advantages in the above three aspects or those that can form cooperative advantages and complement each other in these aspects are expected to stand out in the fierce competition of ADC drug development.
